I. Background
DLL4（Delta-like ligand 4）主要表达于血管内皮细胞，通过 “细胞-茎细胞"分化机制调控血管分支：DLL4 激活邻近细胞的 Notch1 信号，抑制其分化为细胞，从而限制血管过度生长，维持血管网络的有序性。在肿瘤微环境中，DLL4 在异常血管中高表达，诱导血管结构紊乱（渗漏、低灌注），促进缺氧和耐药。此外，DLL4 参与神经发育及 T 细胞分化，其基因突变可导致 Adams-Oliver 综合征等先天性疾病。 DLL4 与 Notch1 是 Notch 信号通路的关键组分，通过细胞间接触依赖性信号调控发育、血管生成及疾病进程。DLL4 是 Notch 配体家族成员，作为跨膜蛋白特异性地结合并激活 Notch1 受体，驱动细胞命运决定、组织稳态及病理重塑。
