| I. Background | |
| 血小板生成素 （TPO） 是巨核细胞生成的主要细胞因子调节因子，通过其特异性细胞 表面受体 MPL 起作用。缺乏 TPO 或 MPL 的小鼠在稳态下产生约 10% 的正常巨核细胞和 血小板，并且在骨髓 （BM） 移植或细胞毒性药物等应激下血小板的恢复减弱。MPL 信号 转导在维持正常的造血干细胞（HSC）数量和活动方面也起着重要作用。除了刺激巨核生 成外，TPO 还可以单独或与其他细胞因子（包括 GM-CSF、IL3、IL6、IL11、SCF、FLT 配 体、FGF 和 EPO）联合支持造血干细胞和 mk 祖细胞的存活和扩增。 TPO 与其特异性受体 Mpl（TPOR） 蛋白结合，触发 Janus 激酶 2 （Jak2） 的激活和 受体的磷酸化，Tpo/Mpl 激活的主要信号级联反应包括 JAK/STAT、PI3K-Akt 和 Ras- MAPK1/3 通路。 | |
