IRA Effector Reporter Cell
| I. Background | |
| 胰岛素受体（INSR，Insulin Receptor）是调控葡萄糖代谢、细胞生长及能量稳态的核 心跨膜受体。INSR 由两个胞外α亚基（配体结合域）和两个跨膜β亚基（激酶活性域）通过 二硫键连接成四聚体（α2β2）。胰岛素结合α亚基后，触发β亚基自磷酸化并激活其激酶活性，进而通过 PI3K-AKT 和 MAPK/ERK 信号通路协调代谢与增殖功能。INSR 作为 代谢与增殖信号的“分子开关"，在糖尿病、癌症及罕见遗传病中具有核心地位。 INSR 通过选择性剪接具有 IRA 和 IRB 两种亚型。IRA 充当胰岛素、胰岛素原和胰岛 素样生长因子 2（IGF2）的受体，介导胚胎和肿瘤组织中的增殖作用，而 IRB 更专化于肝 脏、肌肉和脂肪等代谢组织中的胰岛素作用。IRA 属于激酶受体家族，其胞内段具 有激酶活性，胰岛素结合后触发自磷酸化并激活下游信号。 | |
