I. Background
ENPP3 基因（又称 CD203c 或 NPP3）位于人类染色体 6q23.2-q23.3 区域，编码一种Ⅱ 型跨膜蛋白，属于外核苷酸焦磷酸酶/磷酸二酯酶（ENPP）家族。ENPP3 的核心功能包括水解细胞外核苷酸（如 ATP、GTP、UTP、CTP）和磷酸二酯键，调节嘌呤能信号传导。它通过消除细胞外 ATP（一种触发炎症细胞因子释放的信号分子）来抑制慢性过敏和炎症反应中肥大细胞及嗜碱性粒细胞的活化。此外，ENPP3 还具有磷脂酶 D 样活性，参与磷脂酰的水解，生成磷脂酸，影响膜重塑和囊泡运输等细胞过程。ENPP3 通过其多酶活性和广泛的生物学功能，在骨骼代谢、免疫调节、癌症进展及过敏反应中发挥重要作用，成为具有潜力的治疗靶点。
