I. Background
白细胞介素 22（IL-22）是 IL-10 细胞因子家族的成员，是一种新兴的 CD4+Th 细胞因子，在抗微生物防御、体内平衡和组织修复中发挥重要作用。该家族还包括其他细胞因子如 IL-10、IL-19、IL-20、IL-24、IL-26 和 III 型干扰素（IFNλ）。IL-22 是一种主要由淋巴细胞分泌的细胞因子；包括先天性和适应性免疫系统，例如 3 型固有淋巴细胞（ILC3）、 αβT 细胞、γδT 细胞和 NKT 细胞。在没有明显感染或炎症的情况下，即处于稳定状态，IL- 22 的主要来源是 ILC3，它大量存在于大肠和小肠的黏膜中。在 αβT 细胞中，产生 IL-22 的主要是 Th1、Th17、Th22 细胞；大约 33% 的产生 IL-22 的 CD4+ T 细胞 Th1 细胞，50% 是 Th22 细胞，15% 是 Th17 细胞。中性粒细胞也可产生 IL-22 。功能性 IL-22R 是由 IL-22R1 和 IL-10R2 组成的异二聚体，IL-10R2 与 IL-10 家族的各种细胞因子受体复合物共有。IL-10R2 普遍表达，而 IL-22R1 仅在如支气管、肝脏、胰腺和肠道和基质细胞的表皮细胞中表达。在肝脏中，肝细胞同时表达 IL-10R2 和 IL-22R1；许多疾病，如牛皮癣、移植物抗宿主病、肝脏和胰腺损伤、溃疡性结肠炎和肿瘤与 IL-22 密切相关。在 IL-22 与 IL-22 受体结合后，Jak1/Tyk2 激酶被激活，导致这些受体和转录激活因子 STAT1、STAT3、STAT5 蛋白的磷酸化。STAT3 诱导涉及多种信号通路的许多基因的表达，包括与细胞凋亡、细胞周期等有关的通路。除了 STAT 信号传导外，IL-22-IL-22R1-IL- 10R2 复合物还导致 MAPK 和 p38 通路的激活。
