IL36 Effector Reporter Cell(Adherent)

I. Background
IL-1 家族包括 11 种细胞因子，7 种具有激动剂活性的配体（IL-1α，IL-1β，IL-18，IL- 33，IL-36α，IL-36β，IL-36γ）和 4 种具有拮抗活性的成员[IL-1 受体拮抗剂（IL-1Rα）， IL-36Ra，IL-37，IL-38]。IL-1α，IL-1β，IL-18，IL-33，IL-36α，IL-36β，IL-36γ触发 MPKK 和 NF-κB 导致炎症反应。与 IL-1 家族的其他细胞因子成员类似，IL-36 激动剂（IL-36α、β、γ）与由 IL-36R 和 IL-1R 辅助蛋白（IL-1RAcP）组成的异二聚体受体结合。IL-36R 由许多不同的细胞类型表达，包括角质形成细胞、肺成纤维细胞和上皮细胞、内皮细胞以及各种免疫细胞。IL-36R 在人 M0 和 M2 巨噬细胞中也高表达，但在 M1 巨噬细胞中不高表达。IL-36Rα 和 IL-38 对 IL-36 信号通路具有负向调节作用。IL-36Rα通过与 IL-36R 竞争性结合、抑制 IL-36 激动剂识别和 IL-1RAcP 募集来表现出拮抗功能，从而抑制该家族的激动成员对受体的激活。除了阻断 IL-36R 介导的 MAP 激酶和 NF-κB 通路的激活外，IL-36Ra 也可以阻止 Th17 细胞因子的表达。

II. Description
