IL33 Effector Reporter Cell(Adherent)

I. Background
白细胞介素 33（IL-33）是 IL-1 细胞因子家族的一员。IL-33 与 IL-33R(也称为 ST2)和 IL-1 受体辅助蛋白(IL-1RAcP)形成的异源二聚体结合，启动信号转导，导致局部免疫反应。 IL-33R 在 T 辅助 2 (Th2)细胞、肥大细胞、活化白细胞如调节性 T (Treg)细胞、2 组先天淋巴样细胞(ILC2s)、CD8+ T 细胞以及自然杀伤(NK)细胞中大量表达，从而诱导 Th2 相关细胞因子基因的转录和宿主对病原体的防御。IL-1 受体辅助蛋白(IL-1RAcP)分别是 IL-1Rα、 IL-33R 和 IL-36R 的共受体。 IL-1RAcP 与 IL-33 无直接关联。它被招募到 IL-33R 中，在 IL-33 刺激后形成激活的异二聚体受体复合物。受体复合体然后通过其 TIR 结构域招募下游信号成分。激活的受体复合体随后招募下游信号蛋白，如髓样分化初级反应蛋白 88 (MyD88)、肿瘤坏死因子受体相关因子 6 (TRAF6)，随后诱导 IL1R 相关激酶的激活，如 IRAK-1/4，激活丝裂原活化蛋白激酶(MAPKs)、ERK、p38、JNK 和下游 IKK 复合体，导致转录因子 AP-1 和 NF-κB 的激活。 IL-33 已被确定为多种炎症性疾病的介质，如哮喘、心血管疾病和过敏性疾病等。

II. Description
