I. Background

ICOS 是一种在 T 细胞活化后才被诱导表达的共刺激受体，而 ICOSL 是其配体，主要组成性表达于 B 细胞和抗原呈递细胞表面。两者的核心作用机理在于，当活化的 T 细胞（特别是滤泡辅助性 T 细胞）迁移至淋巴滤泡并与 B 细胞相遇时，ICOS 与 ICOSL 的结合为 T 细胞提供了关键的共刺激信号，这主要通过激活 PI3K 等下游通路，驱动 T 细胞增殖、存活及产生细胞因子（如 IL-21），从而精确调控生发中心反应，最终促进 B 细胞进行抗体亲和力成熟与类别转换，产生高效体液免疫。
