| I. Background | |
| 肿瘤抑制素 M （Oncostatin-M ，OSM）是白细胞介素-6 (IL-6)家族的一种多效性细胞 因子，参与多种炎症反应，如伤口愈合、肝脏再生和骨重塑。除了 OSM，IL-6 家族成员包 括 IL-6、白血病抑制因子(LIF)、IL-11、IL-27、IL31、心营养因子-1 (CT-1)、纤毛神经营养 因子(CNTF)、心营养因子样细胞因子 1 (CLCF1)。OSM 在体内广泛表达，T 细胞、单核/巨 噬细胞、中性粒细胞等多种免疫细胞均可表达 OSM。 IL-6 受体家族的受体复合物均含有 GP130 亚基。OSM 的受体复合物均为异源二聚体， 根据受体复合物中第二亚基的不同，受体复合物可分为 I 型和 II 型两种类型(21)。I 型 OSM 受体复合物由α亚基 GP130 和β亚基 LIFRβ (LIF 受体β亚基)组成，而 II 型 OSM 受体复合物 由α亚基 GP130 和β亚基 OSMRβ (OSM 受体β亚基)组成。当 OSM 和 OSM 受体复合物的两 个亚基同时存在时，OSM 首先与 GP130 形成低亲和力的异源二聚体，然后该异源二聚体 招募 OSMR 或 LIFR 并与之结合，并激活多种信号通路，包括 JAK/STAT、MAPK、JNK 和 PI3K/AKT 通路。 | |
