ERBB Family Effector Reporter Cell

I. Background
      ERBB 家族(又称 HER家族)是一类在细胞生长、分化与存活中至关重要的受体激酶,其异常激活是驱动多种癌症(如肺癌、乳腺癌、胃癌)的关键因素。该家族包含 四名成员:EGFR/HER1、HER2、HER3 和 HER4。它们各自的特点为:EGFR 拥有多种配 体;HER2 无已知直接配体但信号能力强;HER3 激酶活性缺失,必须与其他成员协作; HER4 结构复杂。其中,HER2 是最核心的信号放大器,而 HER3 是关键的信号伙伴, 两者的异源二聚体是驱动肿瘤生长和耐药的主要信号单位。

      ERBB 家族通过配体诱导的二聚化机制启动其核心信号通路。当生长因子(配体)结合后,受体形成同源或异源二聚体(尤其是以 HER2 为中心的异源二聚体),引发胞内激酶区的相互磷酸化。这些磷酸化位点随后募集并激活下游两条主要信号通路: RAS/MAPK 通路(调控基因表达和细胞增殖)与 PI3K/AKT 通路(调控细胞存活和代谢)。这条从膜受体到细胞核的强大信号传导级联,在正常情况下精密调控生理过程,一 旦因受体过表达或突变而失控,便会持续驱动肿瘤的生长、侵袭和生存,这也正是针对 EGFR 和 HER2 等靶点的药物(如单克隆抗体、小分子抑制剂)能够有效治疗癌症的根本原理。
 
II. Description
